This post is funded by Cancer Research UK and will work on a project entitled ‘Exploring the Relationship Between Antigen-Specific T Cells and the Tumour Microenvironment’.
We have evidence that the anti-tumour T cell response can, under certain circumstances, drive structural alterations to the tumour microenvironment (TME) which serve to amplify the immune response leading to tumour destruction. Our findings indicate that changes to the TME are widespread and are characterized by the development of specialised blood vessels named high endothelial venules (HEV), re-organization of lymphatic vessels and loss of extracellular matrix (ECM). Based on these data, we hypothesise that tumour rejection may be improved by equipping particular antigen-specific T cells with the ability to drive alterations in the TME.
We need an experienced molecular biologist to genetically modify a range of in vivo and human antigen-specific T cells already present within the lab. The individual will work with a team of cellular immunologists to test the ability of modified T cells to function in vitro and in vivo. For this purpose, a range of techniques including confocal microscopy, T cell-organoid co-culture systems and in vivo models will be used. The overall purpose of the work is to determine whether T cell-based immunotherapy can be enhanced by empowering adoptively transferred T cells with the ability to alter the TME. The lab is situated at the Henry Wellcome Building, Cardiff University, UK.
