Pulmonary arterial hypertension (PAH) is a fatal disease affecting both humans and animals, for which there is no cure except lung transplantation. The indolent nature of disease means individuals are unaware they have PAH until they present with fully established disease, by which time it is too late. Furthermore, although existing therapies improve exercise capacity in patients, they have limited impact on mortality.
Therefore, there is a major unmet clinical need to develop novel transformative therapies.
We and our collaborators have developed (i) genetic stem models of human pulmonary vascular cells that can be transitioned between disease-free and diseased states and (ii) a new 3D system that allows us to build functional human blood vessel in a dish in the lab (termed biomimetic vessels) that can model pulmonary arteries.
In this project we aim to develop a 3D human induced pluripotent stem cell model of small pulmonary arteries that allows us to understand the process of vascular remodelling that underlies PAH. This project will incorporate genetic engineering of human induced pluripotent stem cells. The success of this project will represent a step change in PAH research and has major advantages over the use of current animal models. It will allow us to identify novel druggable pathways relevant to human disease leading to more effective treatments. This platform will also have significant impact in improving the efficiency of, and reducing the costs of, drug screening, drug development, drug safety testing and clinical trials. Importantly, our work here will also lead to substantial reduction, refinement, and replacement of animals in cardiovascular and cardiopulmonary research.

This PhD studentship is funded by the Humane Research Trust. Funding is available to and applications are welcomed from candidates eligible for home tuition fees only ( ****). Funding comprises home tuition fees and an annual stipend of £15,609 (for a maximum of 36 months).
